    Abstract: This invention discloses means for obtaining immunogenic peptides, polypeptides, proteins, and their corresponding nucleic acid sequences, target cells with vaccine interest, or lysates thereof, without making structural changes in said antigens, through their association with Very Small Size Proteoliposomes. The object of the invention is to provide immunogenic compositions containing peptides, polypeptides, proteins, their corresponding DNA sequences, cells or their lysates and Very Small Size Proteoliposomes (VSSP), which are formed by binding the Outer Membrane Protein Complex (OMPC) of Neisseria meningitidis with gangliosides, by means of hydrophobic links. Additionally, it is stated that these compositions can be formulated alone or in the form of emulsions with the Incomplete Freund's Adjuvant (IFA), and may also be lyophilized.

    Abstract: The present invention describes a pharmaceutical composition whose active ingredient includes conjugates of membrane vesicles of Neisseria meningitidis and the GM3 ganglioside in a conjugation ratio in excess of proteins, has particular characteristics of size, surface charge and a morphology associated with nano-particulate systems that give it advantageous properties as an immunomodulator, because it induces a convenient and significant reduction of myeloid-derived suppressor cells that has an impact on the response of lymphocytes and on the survival of patients with tumors. The invention further discloses the use of the pharmaceutical composition disclosed in the treatment of cancer, particularly those cancers where the myeloid-derived suppressor cells (MDSCs) are high; as well as a method of treatment with said composition in cancer patients and a method to select those who will receive said treatment.

  • Immunopotentiating formulations for vaccinal use

    Abstract: The present invention is related to the field of medicine, particularly to the use of new adjuvant formulations with vaccine antigens. The technical objective pursued with this invention is, precisely, the development of formulations that are able to increase and/or modulate the immune response of the organism to vaccine antigens in the serum and the mucous lining. With this aim, a formulation was developed that contained as the main components the surface antigen of the hepatitis B virus and the acemannan in adequate proportions. As an extension of this result, formulations were developed in which a) HBsAg was used as the hom*ologous or heterologous antigen carrier b) delivery systems of particulated antigens and c) soluble antigens, combined with the acemannan in specific proportions. The formulations of this invention are applicable in the pharmaceutical industry as vaccine formulations for human and veterinary use.

    Abstract: Vaccine compositions in which gangliosides and the OMP of N. meningitidis were combined to form very small size proteoliposomes (VSSP) to be administered subcutaneously are described, these compositions do not require the use of any additional adjuvant. The described compositions allow the immunological treatments with gangliosides, particularly N-AcGM3/VSSP and N-GcGM3/VSSP, showing advantages due to the less aggressive reaction in the site of injection and can be used in a simpler way and better for the patients.

    Abstract: The present invention describes fusion proteins based on cytokines, called bi-cytokines (BC), specifically formed by the binding of an IL2 agonist mutein with a type I interferon (IFN), linked by an Fc region of a mutant human IgG1 and a connector peptide. The combination of an IL2 agonist mutein and a type I IFN in the structure of the bi-cytokines gives surprising immunoregulatory properties to these molecules and a superior therapeutic effect than that of parental cytokines, or their combination, which makes them attractive and novel molecules for the treatment of cancer. Pharmaceutical compositions comprising as an active ingredient the fusion proteins object of this patent are also described.

Depending on the fine structure of the fatty acid in the ceramide of the synthetic GM3, said adjuvants are able to stimulate specifically and in a specialized way the humoral or cellular immune response against accompanying antigens. Particularly, this invention provides immunogenic vaccine compositions that comprise peptides, polypeptides or proteins and the aforementioned nanoparticles, which are formed through the dispersion of hydrophobic proteins of the outer membrane complex (OMC) of Neisseria meningitidis in solutions containing fully synthetic variants of the GM3 ganglioside.

    Abstract: The current invention relates to the field of Biotechnology applied to human health. Here it is described a vaccine vehicle wherein toxins from eukaryotic organisms are encapsulated into multilamellar vesicles obtained by the dehydration-rehydration procedure whose lipidic composition is dipalmitoylphosphatidylcholine:cholesterol in a 1:1 molar ratio for subcutaneous or intramuscular administration. These compositions do not require the use of other adjuvants. The disclosed compositions allow modulation of CTL-specific immune response against one or several antigens co-encapsulated into toxin-containing liposomes. The vaccinal vehicle of the present invention shows advantages over others disclosed by the previous art due to the robustness and functionality of the induced immune response as well as its immunomodulating properties.

The invention describes a vaccine vehicle which toxins from eukaryotic organisms are encapsulated in liposomes with multiple lipid layers, obtained by means of the process of dehydration/rehydration, the lipid composition of which is dipalmitoylphosphatidylcholine:cholesterol in a molar ration of 1:1, which are designed for subcutaneous or intramuscular administration. These compositions do not require the use of other adjuvants. The compositions described allow modulation of the specific CTL immune response to one or more antigens co-encapsulated in the liposomes that containing the toxin. The vaccine vehicle of the present invention presents advantages as compared with others described in the prior art owning to the robust and functional nature of the immune response induced and also the immunomodulating properties thereof.
